This is a mouthful.
Has anyone successfully PCR amplified from DNA that was extracted from
nuclei that was from paraffin-embedded sections? The samples that i have
access to were from paraffin-embedded sections (50micron) of colonic
polyps. Nuclei were prepared from these samples for flow cytometry to
determine ploidy and aliquots were saved for PCR amplification from the
DNA from these nuclei(i have nuclei that were not sorted). I am looking
for a protocol to extract DNA from these nuclei and wonder if anyone has
done this successfully.
