Hi Kam,

neo from Tn5 is itself a very poor choice
because it is an effective silencer
(see Gene, 99, 249).
We use it for YAC transfer into ES cells,
but we have to modify YACs with 2 copies of
the neo-r so that that affective selection
can take place, though again  G418 conc
is titrated very precisely for every batch
of the drug
If tk fails to express you will never know
why: poor orientation or inhibitory
effect of neo-r gene
