Help needed with FISH

I am trying to perform FISH using extended chromatin fibres, using a 
selection of cosmid probes labelled with digoxygenin and biotin. 
However, while I can get the probes working fine on metaphase 
chromosomes, I seem to get either no signal, or just non-specific 
binding on the extended chromatin.
	If anyone can suggest a means to overcome this, or supply me with a 
protocol, I would be most grateful to be emailed the information.
