Kevin Lease asked about PCR of lambda gt10.
My experience is with gt 11 and now, charon 21a. In each case, I haven't
had problems (and in fact have had good success) starting things off with
a 1 minute 94 degree step. Then PCR as usual. These conditions work for
phages and ecolis. I haven't tried them with yeast but may very soon.
