I am not sure. You see, Urea gels have a very high salt conc. (5-8M Urea),
and they polymerize fine, and in fact much faster too (for example the
sequencing gels, IEF gels etc.). It could have been a pH problem. Did you
titrate the buffers to the right pH's? And what did you use - NaOH or
KOH. I do not know if the K/Na salt at high conc. does interfere with
polymerization. Dr. Pillay says he actually uses The base and HCl form
according to the Handerson-Hasselberg eqn. to constitute Tris-Saline
buffer. He did not mention any thing about using the TrisHCl in his
SDS-PAGE.
