Promega's Altered Sites kit

I too have had great success with Promega's Altered Sites kit. I find the 
limiting factor is getting the MutS strain competent enough. 
Electroporation works best for me. However if the mutant phenotype is 
easily screenable (eg via an introduced restriction site) then I 
generally don't bother with the MutS strain and just use our "normal" lab 
E. coli. You still get c. 40% mutants.
