I agree with the previous post...

However, I use Denhardt's Hyb solution....and stick to hyb's and 
prehyb's at 65C....again, I agree dont be too stringent in your 
washes...

check the blots after each wash...

start at RT in 0.5xssc/0.1%sds then move to
40C in 0.2xssc/0.1%sds

and then to 

55C in 0.1xssc/0.1%sds

check the blots at each step....if the counts get too low then dont wash 
any more....
