I did a plaque lift from plasmid-transformed E.coli, hybridized them and got some positive colonies. The problem is all my colonies died and I didn't do replica. My question is: is there anyway of recovering the plasmid DNA from the membrane (Hybond N+) (which I have already dried). Any suggestion will be appreciated. Magda
