pWE15 screen

Hi all!

I'm about to begin screening a genomic library in pWE15 (a cosmid) for a
gene I think exists as a single copy.  Does anyone have experience in 
this sort of procedure?  I do not want to spend the next three years doing
this so any tips on what to try and what absolutely to avoid would be welcome.
I have looked into Maniatis and have the screening protocol from Stratagene.  
The prospect is a little daunting, since I haven't done this before, so any 
help would be appreciated.
