I have found that I have had the best results using that Fluoro-dATP works best when the following conditions have been met: 1. Somewhere in the next 8-10 bases there is one (and preferably only one) incorporation site is present. 2. The primer that is used does not form a primer dimer. 3. The primer is allowed to anneal specifically to a single site.
