Michael,

I modified the method of Horton, et al. (1990) Biotechniques 8, 528-535.
Rather than join two unlike cDNA's like the method described in the paper,
I use the general principles to generate mutants in my target cDNA.  So
far, I have not failed to create my specific mutant.  A lab next door uses
Pfu polymerase for these procedures.  I have found that Taq works fine.
