Subcloning large fragments

about subcloning large fragments of DNA :

- did you already used other strans than XL1 blue !

- do you transform your cells by electroporation or heat shock ?

- how don you prepare your cells for transformation ?


I have many answers for those who are interested in transformation,
but I have many problems with the low efficacity of my transformation

I think that my cells are not well prepared
