Good question. Our lab switched completely from columns to a manual
method for exactly this reason. Called Qiagen a couple of times but
they told me it needed to be optimized for each plasmid. we had preps
from the same constructs giving us completely different yields. The
DNA was fine, but this way it became quite expensive.
