Hello -
Has anyone out there used a protocol written up by Souer, et al.,
(from "A general method to isolate genes tagged by a high copy number
transposable element", E. Souer, et al, The Plant Journal, 1995
7:677-685)? It uses a combination of inverse polymerase chain reaction
and differential screening of amplification products to isolate
transposable element flanking sequences unique for mutant plants. It also
uses a "vectorette" system - anyways, it sounds really slick, and would
help to avoid a million Southerns, but there are a couple holes in the
protocol (can't get a hold of the folks, are at Vrije Universiteit,
Amsterdam).
