Is it genomic DNA that you want or do you want mRNA for cDNA library
construction? 
Again eukaryotes are quite a variable group as far as cell disruption goes
and some organisms such as nematodes for example require special tricks to
get HMW DNA out. Some general clues would be very heplful.
