I hate CIP! It does damage ends, and takes ardous deactivating. I use SAP (from USB, now Amersham I expect) and find it much friendlier. Running your ligations out on a minigel is good advice, but I prefer 1:1 molar ratios to insert biased ones (call me a purist). Anthony.
