Shaking is definitely not necessary. If you use hybridization buffer with
50% formamide, it suffices to simply moisten each filter (for colony
lifts, for example,1--2 ml buffer per filter; to be sure that all filters
are evenly wet, put one filter at a time into a Petri dish and cover with
aliquot of buffer before adding the next filter.)
