transcriptional activity and mRNA stability

Dear Osama,

Nuclear run-ons will measure transcriptional activity, but mRNA 
stability must be addressed using Actinomycin D (an RNA Pol II 
inhibitor).  I don't have a protocol handy, but basically you incubate 
the cells of interest with Act D and prepare RNA for Northern analysis 
at various time points.  The decrease in signal over time is a measure 
of your mRNA degradation, since no new transcripts are being made during 
the incubation.
