T7 RNA Pol in vivo assay

Hi netters!!!!!!

	Finally I have my mutations of the T7 RNA pol, I need to assay the 
activity of those. First I need to found an active enzyme. I read 
that rimfamicin inhibits the RNA pol of E. coli and of curse I can use 
crude extracts with template for mesuare if is or is not an specific 
mutant active. Does anybody have done this kind of experiments before??? 
and could give and advice or tips for that seleccion??
