Your basic stupid question about PCR cloning

The one thing no one has mentioned is how to get it in. Never mind which
vector. There have been a few posts over the last year where people have
had no success at all cloning these large fragments. If you have unique
internal sites you may be better cloning it in bits and then trying to
stick it back together again. 

Please let us know how succesful or not you are.
