Hiya,
I have often used a colony prep protocol which involves boiling an E.coli
colony picked into a small volume of TE for 10 minutes. This lyses the
bugs, and i presume it would denature the plasmids released as well.
However i then went on to transform some of this colony prep back into
E.coli, and this worked fine (can't remember the numbers, but it was a
lot). All i had done with the colony preps after heating was pop them in
an ice bucket, so i think it is just a case of heating up and cooling
down, and they will anneal.
