if you think that it's a problem with changing annealing temps, why don't
you try a touchdown pcr program. check out BioTechniques 20:478-485
(mar.96) for an example program. we use a touchdown program of our own
for a whole mess of 33p 5' end-labelled microsatellite primers, no matter
the annealing temperature, and it works great!
