One big plus for the CAT assay method which involves partition of 
labelled product into scintillant (Neumann et al BioTechniques 5, 444
-447 (1987); Eastman ibid 730-732) is that you can take data from a 
reaction over a time course and hence establish the initial rate of 
conversion of substrate. Only this is a true reflection of the amount
of CAT enzyme present, and since there are enough other assumptions
involved in using an amount of enzyme as a measure of the transcrip
tional activity of the promoter driving its synthesis, it is good to
get the amount of enzyme right.
