If you don't require extremely pure DNA, you could try boiling a few Rhizobium
colonies in 500microlitres of UHQ for ten minutes. Thereafter centrifuge at
around 5000g for 3 minutes. Your DNA will be in the supernatant. I have used
this method successfully to extract Clostridium difficile.
