Insertional inactivation of genes

Hello All,

I would like input regarding how I might insertionally inactivate or
delete a region of genomic DNA, from a cyanobacterium, that exists in an
operon. The antibiotic resistance cassettes I am using now, have within
them transcriptional and translational stop sequences. I want to disrupt
upstream sequences without disrupting the expression of downstream
sequences.

Any advice or input would be greatly appreciated.

Thanks
