Simple- just equilibrate your desalting column with the buffer that you want
your enzyme to end up in. Most of these columns have a MW cutoff of about
5000, so glycerol won't be separated out. Keep in mind that sample size makes
a big difference in the results, however. If you push the limit of column
capacity (about 25% of gel volume for Sephadex G25) you remove only about 90%
of the low MW material.
