Unlike most coupling chemistries, the aldehyde you are using
works well under a broad range of pH conditions. I would suggest coupling at
neutral or even slightly slightly acid pH. Varying the salt concentration may
help also. BTW- you might want to consider coupling in the presence of ligand
in order to protect the binding site.
