5% dry milk in PBS with 0.1% Tween-20 is a sufficient block. For
chemical detection overnight in the cold is OK but for ECL it will give
you that background. You shouldn't need to block more than 20 minutes at
room temp. Of course you then have to do the entire procedure in one
day, but you will think its worth it when you see the result!
