Touchdown pcr

HI

I even found a better protocol that's derived from touchdown but much 
more simple to program and more robust (one program amplifies it all!): 
stepdown PCR (Hecker, 1996; BioTechniques 20:478-485). I am now usiong 
it on a regular basis and am perfectly satisfied with it!
