Sonication Protocol!!

Hi..All.
   I am trying to disrupt the cell, E.coli carrying plasmid DNA.
   We have sonicator in the lab but I am not sure of the settings for
   disruption.  I am guessing that before you sonicate you have to 
   centrifuge the E.coli in microtube and resuspend them in buffer.
   Then, sonicate.....:)

   Well, I'd be appreciated if you give me a piece of clue.
