CoT curves? Genome size determination

I want determine the size of a nematode genome. I was thinking about
using the "classical" biochemical method - a CoT curve. Is there
anything more clever/sophisticated/accurate/easy/cheap? Maybe someone
could point out to me the most updated CoT methodology?
