Hi, I need some advice, could you help me? I have to amplify a fragment of 86 bp. I have two options: 1) Design two primers of about 20-24 bases and run a PCR. or 2) Design two oligonucleotides of 48 bp that overlap in 9 bases and do an extension with a polymerase. Do you have any advices or experiences about this? Thanks, Paola
