Help! chloroplast isolation

If this alga is unicellular then try the chloroplast isolation protocol used
for cell wall minus chlamydomonas published several years ago in Plant Phys. (I
don't remember exact year). Essentially, the cells are pushed through a 27
gauge needle on a syringe. Presumably, the shear forces break open the cells
while the chloroplasts come through undamaged. Then the chloroplasts can be
further purified on a percoll gradient to isolate them away from other
organelles.
