messy agarose bands

One thing we know makes a big difference to the sharpness of bands in 
agarose gels (and acrylamide gels) is the composition of the loading 

There are n recipes around for these using all sorts of "density 
enhancers" ficoll, glycerol, DMSO etc

Years ago we tried 'em all, head to head, and by far the best in our 
hands was sucrose.  We make a 40% sucrose / 200mM EDTA/ bromophenol 
blue, stock (store at 4oC for years!) and add 1/10 V/V to restriction 
digests, fragments, etc.

Try it and see if that helps.
