I AM NOT MUCH OF AN EXPERT ON PROTEIN EXPRESSION BUT HAVE DONE SOME
EXPRESION USING pET vectors. IF YOUR PROTEIN IS TOXIC THEN USE A HOST
WITH STRINGENT INDUCTION CONTROL SUCH AS BL21DE3 pLYS S and a vector
with an inducible promoter such pLAC. This would give you 2 levels of
control. First the promoter (IPTG induced) second the pLYS S that binds
to the T7 RNA polymerase if it is made in small quantities. Hope this
