A simple method to quantify peptides is by means of HPLC at 214 nm. The
signal here is nearly proportinal to the
number of peptide bonds. So what you need is one accurate standard (short
peptide). Weighing is not as inaccurate as
you think. The yields are usually around 70 to 75 percent.
