i agree with dima - the salt is the problem
we process the ligations thus:-
add 2 ul strataclean resin vortex, spin.
transfer the solution to a whatman microdialysis membrane floating
on a petri dish of water. leave for an hour or so - transfer to
fresh tube for electroporation.
this works every time - no arcing and better frequencies than with
precipitated dna (we did a head to head experiment to test this)
