Hello, we are cycle sequencing with Thermosequenase from amersham, and there is one stretch that is impossible to read with any primer pair that we use. we do not know whether it is just CCG or CCGG or CCCGG. the sequence surrounding this problemspot is not very GC rich. Any suggestions ? can one use DMSO, and at what concentration ? z
