I am looking into the expression of phenylpropanoid genes - PAL, COMT
and CCOMT- specifically involved in the lignin synthesis, in the wheat
cultivars resistant and susceptible to a leaf spot disease, using
alfalfa phenylpropanoid cDNA clones- CCOMT, COMT and Rice -PAL cDNA
clone as probes.
My question is - since the homologies between Alfalfa (Dicot) and wheat
(Monocot) aren't that great, what are the conditions I would have to
consider while performing Northern hybridisation?
I would like to perform the hyb. with total RNA isolated from the
Any suggestions/comments welcome.
