delete signal peptide ?

Hi,  all. I plan to make a eukaryotic protein in E coil. This protein  has
a signal peptide. Before starting my construction, I want to know if I
need to delete this signal sequence form my construction? Does it interfer
the folding process of mature protein and where will this protein be ( in 
culture medium, membrane or  cytosol ?) if this signal peptide is included
in the construction?
