Hi newsgroupers, we have tremendeous problems in cloning a retroviral vector (about 10 kb) in E.coli C600 or HB101. Our bacteria grow very good in LB-amp but the plasmid yield is extremely low. We tried Qiagens as well as Machery and Nagels midi and maxi plasmid prep kits. Any suggestion is appreciated. Thanks a lot Roger
