We have been having some trouble here in our lab with Agrobacterium
tumefasciens strains used for Arabidopsis transformation.  They seem to
lose the Agro part, even when stored at -80, and are then just E. coli
leftovers from the triparental matings.
This is determined using a ketolactose test.
Are there any better tests than this?
Is it possible that the E. coli strains used for tri-parental mating
have somehow gained resistance?
