It is a method to look at unknown sequence adjacent to a known region. DNA
can digested with a RE within your known sequence and religated to unknown
sites to form closed circular molecules. PCR is used with two oligos
containing 3' ends facing away from the 5' ends (the opposite way of normal
PCR). If your lucky and have chosen a RE that generates a reasonable size
circular DNA you can amplify neighboring sequence for cloning or
manipulation.
