Uracil containing DNA and Proof-reading pols

Hi Folks,

There have been a few posts recently about the use of Pfu or Vent etc
with uracil containing primers or using dUTP in with the nucleotides. As
I just came across a paper in my filing cabinet about this it may be of
interest.

Lasken et al J.Biol.Chem Vol 271 No. 30, pp17692-17696, (1996)

The work was done by Life Technologies and basically shows that  uaracil
containing DNA binds with 6500 fold greater infinity than non-uracil
containing DNA so effectively acts as an inhibitor. dUTP as a nucleotide
was incorporated with approx 40% reduced rate.
