Dear Neters I want cloning a gene promoter from Arabidopsis. I know the sequence. And this gene is the single copy. the promoter region is about 1100bp long. I am not sure whether I should direct PCR this promoter(using one pair of primers) or Nested PCR (using two pairs of primers). Anybody has the experiences on this aspect is welcome. Ma
