Ladies and gentlemen,

I'd like very much to take part in the method's group.

I have now a problem to amplify some 4 kb fragment
from RT-PCR. What I am using is ULTma Polymerase,
Perkin Elmer, with the recommended Hot Start technique,
facilitated by AmpliWax Gem 100, Perkin Elmer. I have
tried different annealing temperatures, as well as
different concerntrations of MgCl2 and dNTPs, but never
got a correct band, although the templet seemed to be
OK. Should I use some help system, or should I just
change the polymerase?
