Cloning Vector effect ?

Hi Netters,

I have a unique problem. I cloned a 5 kb piece of DNA in pUC18 and the
same one in pZErO vector. I get deletions in the insert cloned in pZErO
but not in the one cloned in pUC18. I would prefer to use pZErO and not
get any deletions. Did anyone else observe this or have a clue to what
might be happening ? I appreciate any comments/suggestions.
