We typically use 10ul of 35S-UTP (10mCi/ml, Amersham) in a total reaction volume of 30 ul with good result. The amount of DNA template in our reactions is closer to 500 or 600ng than to 100ng. Best of luck. Ed Frank Baylor College of Medicine
