the difference between vaccinia virus topI and the mammalian topI is its
sequence specificity for 5'-(C/T)CCTT-3' with topI being covalently linked
to the 3'-T. mammalian topI cleavage is almost unspecific.
Another question: is the plasmid in the cloning kit already activated with
topI?
