PCR cloning problems

This happens all the time. What you should do is always run a few controls after
you have got a PCR product before further cloning or sequencing. You should do a
few PCR with only one single primer (forward or reverse) and two primers (F and
R). If you get the product with either one primer, then forget about it. Start
all over again. This could save you doing all the work afterwards. Good luck and
hope your boss doesn't kill you.
